So What is the Ras/MAPK pathway

On September 10th 2010, we will be launching a new network in the UK, the RASopathies Network. The launch is being held at the house of Lords in front of an invited audience of health professionals, and families effected by one of the syndromes that make up this class of genetic disorders. So what are ‘RASopathies’.

Recently a class of developmental syndromes, the ‘RASopathies’, is caused by germline mutations in genes that encode protein components of the critical signal transduction cascade, the Ras/mitogen activated protein kinase (MAPK) pathway. At present, these syndromes include:-

  1. Noonan syndrome
  2. LEOPARD syndrome
  3. Hereditary Gingival fibromatosis
  4. Capillary malformation-AV malformation syndrome
  5. Neurofibromatosis type 1
  6. Legius syndrome
  7. Costello syndrom
  8. Cardio-facio-cutaneous syndrome
  9. Autoimmune lymphoproliferative syndrome

The Ras/MAPK pathway is essential in the regulation of the cell cycle, differentiation, growth and cell senescence, all of which are critical to normal development. Therefore, it is not surprising that its dysregulation has profound effects on development. Each syndrome is this calls of developmental syndromes, exhibits unique phenotypic features, however, since they all cause dysregulation of the Ras/MAPK pathway, there are numerous overlapping phenotypic features among the syndromes, including characteristic facial features, cardiac defects, cutaneous abnormalities, neurocognitive delay and a predisposition to malignancies.

That is a LOT of syndromes, and collectively around 1 in 1000 people will be effected by one of these syndromes.

Understanding this pathway is very important for the simple reason that through the understanding of the RASopathies provide  a valuable insight into normal human development, including many aspects, of Cancer, and other human disorders .

The Jeremy Kyle show

dscf1401 Last week we went to the ITV studios in Manchester to appear on the Jeremy Kyle show.  The staff on the show looked after us well, and JK although very busy was great. The show was about exceptional children.  Helaina was amazing, she stole the show, answering all of the questions like the professional that she is. Full of confidence and shined, though our nervousness was evident.

We do not have a broadcast date yet, it should be on air sometime Early July.

We were honoured to be asked to do the show, especially as we move towards the first UK conference, if one or two families who have no access to the web see the show and realise that there are others facing the same issues they are, and that there is support for them that will have made it all the more worth while. That said telling Helaina’s story has since the recording had an even bigger impact on my life. For the first time in many years I have felt a weight has been lifted, for the past few nights I have slept well, and deeply. Our story told the past now firmly behind us, we can look to the future and the joys that it will bring.

This week is  britains got talent week, bands and singers compete to impress Simon Cowell, sadly many of them are lacking in true talent. There are one or two gems but most of the music is just the same old material re-packaged.

Music has potential to improve communication skills and enhance well being, so why is today’s music just so boring, and when was the last time you listened to something that made the hair on the back of your neck prick up with joy.

So to counter the boring from 2010, please sit back and enjoy the   amazing Nina Simon  recorded live in Montreal, 92,  as she sings I put a spell on you,  pure genius. And why this song out of all the amazing music Nina Simon ever sang… Its simple CS puts a spell………….

With the days flying by I would like to remind everybody who is interested in attending the London conference to submit your final registration as soon as possible.
